Erfolgreiche Teilnahmen am Symposium zur Molekularbiologie der Pilze 2022

Im September 2022 fand das 14. Symposium zur Molekularbiologie der Pilze nach langer COVID19-Pandemiepause gemeinsam mit der GenAG Pilzgenetik der Gesellschaft für Genetik statt. Die Organisatoren rund um Matthias Hahn von der TU Kaiserslautern haben ein vielfältiges Programm mit internationalen Sprechern aufgestellt, die verschiedenen Aspekte der aktuellen Pilzforschung beleuchtet haben. Für unsere Abteilung gab es zwei Höhepunkte: zum einen den Vortrag von Lucas Hollstein über 'Establishment of in vivo protein-proximity labeling with biotin in Sordaria macrospora', zum anderen der Preis an Anina Rudolph für ihr Poster 'Spore types-specific infection of different maize tissues by Colletotrichum graminicola'.

Aktuelle Publikation in Scientific Reports: Establishment of in vivo proximity labeling with biotin using TurboID in the flamentous fungus Sordaria macrospora

Proximity-dependent biotin identifcation (BioID) has emerged as a powerful methodology to identify proteins co-localizing with a given bait protein in vivo. The approach has been established in animal cells, plants and yeast but not yet in flamentous fungi. BioID relies on promiscuous biotin ligases fused to bait proteins to covalently label neighboring proteins with biotin. Biotinylated proteins are specifcally enriched through biotin affnity capture from denatured cell lysates and subsequently identifed and quantifed with liquid chromatography-mass spectrometry (LC–MS). In contrast to many other affnity capture approaches for studying protein–protein interactions, BioID does not rely on physical protein–protein binding within native cell lysates. This feature allows the identifcation of protein proximities of weak or transient and dynamic nature. Here, we demonstrate the application of BioID for the fungal model organism Sordaria macrospora (Sm) using the example of the STRIPAK complex interactor 1 (SCI1) of the well-characterized striatin-interacting phosphatase and kinase (SmSTRIPAK) complex as proof of concept. For the establishment of BioID in S. macrospora, a codon-optimized TurboID biotin ligase was fused to SCI1. Biotin capture of the known SmSTRIPAK components PRO11, SmMOB3, PRO22 and SmPP2Ac1 demonstrates the successful BioID application in S. macrospora. BioID proximity labeling approaches will provide a powerful proteomics tool for fungal biologists.

Forschung - Sordaria macrospora und Colletotrichum graminicola

Schimmelpilze sind hervorragene Modelle um grundlegende eukaryotische Prozesse zu untersuchen. Wir verwenden den Saprophyten Sordaria macrospora und das Pflanzenpathogen Colletotrichum graminicola um unterschiedliche Aspekte genauer zu beleuchten.

Die Aufklärung der molekularen Ursachen von Entwicklungsvorgängen ist ein zentrales Anliegen unserer Forschung. Im Vordergrund steht dabei die Untersuchung der molekularen Grundlagen der Morphogenese komplexer mehrzelliger Strukturen bei Ascomyceten. Diese bilden im Verlaufe ihrer sexuellen Fortpflanzung aus verschiedenen Zelltypen bestehende, komplexe räumliche Strukturen, die sogenannten Fruchtkörper, aus. Colletotrichum graminicola
Kommunikation ist essentiell - sowohl für die Abstimmung bestimmter Prozesse innerhalb eines Organismus, als auch für seine Anpassung an äußere Lebensbedingungen. Wir verwenden das pilzliche Maispathogen Colletotrichum graminicola um sowohl Selbstkommunikation (Keimlingsfusion, Mating) als auch der Fremdkommunikation (Infektion, gerichtetes Wachstum zum Wirt) zu untersuchen. Hierfür verwenden wir genetische, biochemische, bioanalytische und mikroskopische Methoden um Signalstoffe, entsprechende Rezeptoren und die zugehörigen Signalwege zu identifizieren und mögliche neue Anwendungen für den Pflanzenschutz zu entwickeln. mehr...
